Grind lies with pestle make sure all big pieces are broken. Add 125 ul of sb+ into the same tube grind for 10 seconds. Add another 125 ul of sb+ to rinse debris of pestle (total amount of sb+=250) If ly tissue on walls of tube use yellow pipettor to push it into sb+ completely. Complete same steps as above but use the sb- bufer. Place tubes into 60 degree water bath for 25 minutes. Centrifuge 1. 5ml centrifuge tubes (tubes in water bath) for 3 minutes at 14000rpm. Use p200 yellow tip to transfer 200 ul of the supernatant(centrifuged test tube) into a new 1. 5ml centrifuge tube **make note not to take bottom/top oily debris ** Add (18 ul) 5m of nacl into the new centrifuge tube.
