Grew e. coli cells for multiple generations using 15nh4 as the sole nitrogen source to uniformly label the cellular dna with 15n. Increase in mass is used to separate [15n]dna from [14n]dna in a density gradient. 15n-labeled e. coli cells were transferred to unlabeled 14n medium and allowed one round of replication then analyzed. Conservative replication: two bands of duplex dna ( one 15n and one 14n)
