BIOL 461 Lecture Notes - Lecture 12: Cyclin-Dependent Kinase 1, S Phase, Wild Type
Document Summary
Researcher controls when/where a mutant phenotype is displayed. Review: in what kind of situations would you want a conditional mutant: a lethal mutation (essential gene) and you want to get around the lethality. A mutant is at one condition (permissive condition) wild type. Powerful tool used to map pathways of many essential genes (generally to warmer) to a temperature sensitive phenotype, and the restricted temperature, the mutant phenotype is displayed. Third on list of how to look at essential genes/lethal. Phenotypes: conditional mutants: get the organism alive at the permissive temperature and then shift to the restrictive condition/temperature and then in your living organism see what happens. At 25 degrees you have a functional cdk that controls the start cycle (s-phase promoting factor that controls the cell cycle) At 25 this is functioning, and colonies of yeast will form. At 25 degrees the mutant cdk, although it has lower affinity. Shift this mutant to 36 degrees, wt nothing happens.
