Objectives of this section: how the cell synthesizes components of the membrane, the cell ensures that. Endomembrane system: endoplasmic reticulum, golgi complex, endosomes, lysosomes, plasma membrane. *all these 5 are part of something we call the biosynthetic pathway; synthesis and sorting + secretion is their main goal. Rough er = due to presence of ribosome. Rough er: protein secretion, such as: lumen proteins or membrane proteins (made more specifically by the ribosome). Technique: cell fractionation: chop tissue and put into a homogenizer. On ice: centrifuge multiple times, various settings (starting low speed, less time). This divides the different parts of the cells or unfractured cells. Purpose: to separate out (isolate) the different parts of the cell. Need: some type of tissue (like brain, liver . ) or cultured cells. *we will not consider the nuclear membrane to be part of the endomembrane system.
