Nxf/nxt factors ensure that only good rnas are exported out of the nucleus - by binding to ref (mark of splicing factors) and by binding to npl3 (poly(a) marker) This pathway was found out through a genetic screen involving the lacz operon in yeast: lacz encodes for beta galactosidase ; enzyme which makes the colonies of yeast turn either blue when a certain substrate is added. Alternatively, when the gene is not encoded, the colonies turn white: they introduced an artificial intron in the orf. > site-directed mutagenesis: they didn"t change the codon sequence. Wt > normally, the intron gets spliced out white colonies if it is not spliced out, it isn"t exported and white colonies. By mutating mlp1 > this means that the intron doesn"t get properly spliced but the protein is still able to be exported. Mutagenesis : throw chemicals to induce random mutants. They found these cases of blue colonies when that mlp1 protein is mutated.
