Genetic screen: create genetic mutations at random to whole genome and look through the mutations which produced phenotypes of interest (ex: messed up cell cycle) Temperature sensitive (ts) mutant: mutated gene codes for a protein variant which has a restrictive temp (24 degrees) and a permissive temp (37 degrees) so that its function can be turned on or off with ease. Cell division cycle mutants = cdc mutants. There are 2 phenotypes when there are cdc mutants in s. pombe: Elongated phenotype (cdc2-): the cell is longer because g2 stays on and it keeps growing it is a recessive (loss of function mutation) called cdc2- Wee phenotype (cdc2d): the cell is small because it prematurely goes into mitosis without fully growing it is a dominant (gain of function mutation) of the cdc2 gene called cdc2d. Low cdc2 protein the cell fails to divide since it elongates and remains in g2 (loss of function)
