In the lab they made 2 types of dna: one melthylated at v strand, one methylated at c strnd. In 2 sets of different reactions they had their different dnas methylated on c strand and methylated on v strand: these were treated with different enzymes (pink, when detected on te agarose after being treated. If the mismatch can distinguish old methylated from new unmethylated, only the new strand would be nicked: to distinguish this, the agarose gel was analyzed by the radioactive dna probes. If the strands are cleaved it"s denoted by the presence of the h (blue) Initiated but not completed: can have a dsb, c) fork regression, dangerous if not repaired. It wont stick to blunt ends: 3" overhangs need to be around to do the invasion, 2) invasion, 3" end opens up and invades the matching sequence at the bottom, dna is pretty stable. It encounters a blocking lesion and the fork will stall.
