Hematopoeitin family of cytokine receptors are noncovalently associated with 2 protein kinases (jak) Dimerisation of receptors -> jaks come into close proximity. Will phosphorylate the other jak on tyrosine residue. Each jak will then phosphorylate their receptors on specific tyrosine residues. This is recognised by sh2 domains on other proteins. Reside in the cytoplasm in the inactive form. Each stat recognises different sequences of the phosphotyrosines on the receptor, and are thus specific to a certain receptor. Stat will be activated by jak when it binds to the receptor. Ex. protein inhibitors of activated stat (pias) promote degradation of receptors and pathway components. Recognition of different pathogens = different tlrs activated. Results in variation of the cytokines expressed by the stimulated macrophage. Macrophages/dcs secrete il-1b, il-6, il-12, tnf-a and cxcl8. Chemokines secreted will induce chemotaxis in responsive cells. Recruit monocytes, neutrophils, to the site of infe(cid:272)tion.
